Role Summary

We are seeking a highly motivated Scientist I in the Preclinical Sciences team to support our iPSC and primary immune cell engineering initiatives. This position will be responsible for implementing and optimizing a cell engineering workflow from designing and screening appropriate gene editing tools, identifying delivery methods, building medium/high throughput screening in primary immune cells, and the assessment and quantification of on and off-target editing. This role will also play a vital part in supporting Notch’s cutting-edge innovation of therapeutic cell design through molecular cloning and screening of functional payloads, and -omics-based characterization of engineered cells. Direct experience in molecular biology, genomics, and the ability to independently design and execute complex cell engineering experiments are essential.

Key responsibilities:

Support advancement of our genome editing pipeline and platform

  • Screen and characterize gRNAs for gene KO/KI efforts
  • Design, clone, and evaluate nucleic acid reagents for CRISPR-mediated site-specific insertion of transgenes
  • Establish a medium-high throughput genome engineering workflow for gene KO and KI in primary immune cells
  • Carry out appropriate methods for measuring genome editing efficiencies and characterizing repair outcomes for on and off-target editing

Design and implement genomic and transcriptomic NGS assays to characterize engineered cells

  • Execute library design, preparation, QC, and troubleshooting
  • Manage relationships with CROs
  • Collaborate with the bioinformatics team to facilitate efficient data processing and analysis

Participate in the design, rapid construction, and efficient screening of functional payloads for engineered immune cells

  • Collaborate with cancer biology and cell engineering teams to progress designs from conception through various stages of evaluation

Maintain project timeline and report progress regularly to the project team

  • Maintain excellent records of experiments in lab notebooks in a timely manner

Other responsibilities as required

  • Present data at group and company meetings and externally as required
  • Participate in external collaborations with partners and CROs
  • Support lab operations and equipment purchasing/ vendor selection

Role Requirements and Qualifications

  • PhD in molecular biology, cell biology, bioengineering, chemical engineering or a closely related discipline with 2+ years of postgraduate work experience in a biotech or industry setting or master’s degree with equivalent experience in a biotech/pharma setting
  • Extensive knowledge of gene-editing technologies and methods for both KO and KI
  • Proficiency in construct design, codon optimization, and molecular cloning techniques
  • Experience in characterizing cells at the genetic and transcriptional levels using -omics approaches as well as ddPCR and RT-PCR
  • Expertise in execution of NGS wet-lab methods and data analysis using python or R
  • Experience with single cell sequencing and 10X platform
  • Working knowledge of T-cell culture, activation/expansion, and functional assays
  • Experience with flow cytometry for cell analysis and sorting
  • Ability to effectively prioritize, plan, and execute next steps based on experimental results in a fast-paced research environment.
  • Team-oriented, can-do attitude. Motivated to contribute and to learn new techniques.
  • Strong organizational, record-keeping and communication skills, both verbal and written

Desired Characteristics and Demonstrated Skills

  • Immunology background would be an advantage
  • Experience using long-read sequencing technologies (e.g. nanopore, PacBio)
  • Experience working with iPSCs would be an advantage
  • Demonstrated ability and keen interest in modelling Notch’s Core Values: Collaborative, Motivated, Ingenious

Salary Range: $106,000 - $138,000
